Background InformationLeucine-rich repeat-containing G-protein coupled receptor 4 (UniProt: Q9BXB1; also known as G-protein coupled receptor 48, LGR4) is encoded by the LGR4 (also known as GPR48) gene (Gene ID: 55366) in human. LGR4 is a multi-pass membrane glycoprotein that is synthesized with a signal peptide (aa 1-24), which is subsequently cleaved off to generate the mature form that contains four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. LGR4 contains fifteen LRR repeats. It is expressed in multiple steroidogenic tissues. Its expression has been reported in basal and granular cells in the skin, ductal epithelial cells in the breast, spermatogonia and tubule cells in the testis, islet cells in the pancreas, and tumor cells in the majority of colon cancer cases. LGR4 functions mainly through Wnt/β-catenin signaling to regulate cell proliferation, differentiation, and adult stem cell homeostasis. It serves as a receptor for R-spondins that potentiates the canonical Wnt signaling pathway and is involved in the formation of various organs. Upon binding to R-spondins, it is shown to associate with phosphorylated LRP6 and frizzled receptors that are activated by extracellular Wnt receptors. This triggers the canonical Wnt signaling pathway to increase expression of target genes. In contrast to classical G-protein coupled receptors, it does not activate heterotrimeric G-proteins to transduce the signal. LGR4 also acts as a negative regulator of innate immunity by inhibiting TLR2/TLR4 associated pattern-recognition and proinflammatory cytokine production. It is also implicated in the survival of stem cells in the gut. Haplotype insufficiency of LGR4 is linked to increased risk of squamous cell carcinoma of the skin and to reduced birth weight and electrolyte imbalance. Mutations in LGR4 gene are known to cause osteoporosis. Clone 5A3 is reported to detect LGR4 in human and mouse tissues. (Ref.: Yi, J., et al. (2013). PLoS ONE. 8(10); e78144; Li, Z., et al. 2015).
