Background InformationKallistatin (UniProt: P29622; also known as Kallikrein inhibitor, Peptidase inhibitor 4, PI-4, Serpin A4) is encoded by the SERPINA4 (also known as KST, PI4) gene (Gene ID: 5267) in human. Kallistatin, a negative acute phase plasma protein of the serpin family of proteins that displays antioxidant and anti-inflammatory properties. It is expressed in liver, kidney, and blood vessels. It is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off in the mature form. It binds strongly to tissue kallikrein and inhibits its amidolytic and kininogenase activities. This Inhibition is achieved by formation of an equimolar, heat- and SDS-stable complex between the inhibitor and the enzyme, and generation of a small C-terminal fragment of the inhibitor due to cleavage at the reactive site by tissue kallikrein. Presence of heparin can block kallistatin s complex formation with tissue kallikrein and abolish its inhibitory effect. Kallistatin expression is shown to be negatively regulated by oxidative stress via JNK-dependent FOXO1 activation in cultured endothelial cells. Kallistatin is reported to block, via its heparin-binding site, various signaling pathways mediated by growth factors and cytokines, including VEGF, TNF-alpha, TGF-beta and EGF. Kallistatin levels are shown to diminish markedly in rodents with hypertension, sepsis, streptozotocin-induced diabetes, and cardiac and renal injury and its levels are reported to increase in human subjects with rheumatoid arthritis.
