Background InformationInhibitor of growth protein 5 (UniProt Q8WYH8; also known as p28ING5) is encoded by the ING5 gene (Gene ID 84289) in human. ING5 is a member of the ING family of proteins known for their antiporliferation activity. ING proteins bind to methylated core histones through their PHD motif (a.a. 186-235 in ING5), and ING5 is known to target histone H3 on di- or trimethylated Lys4 (H3K4me2/3). ING5 associates with two histone acetylase (HAT) complexes, HBO1 and nucleosomal H3-specific MOZ/MORF, that interact with the MCM helicase and are essential for DNA replication during S phase. ING5 deregulations are reported in various cancers and ING5 located chromosome region is deleted in oral squamous cell carcinoma and odontogenic tumors. In addition, ING5 cytosolic localization is reported in some head and neck squamous cell carcinoma tumors and the cytosolic localization positively correlates with the progression of gastric cancer. ING5 also pormotes p53-dependent gene transcription by enhancing p53 acetylation in a p300-dependent manner. ING5 is shown to interact with p63 that mediates the self-renewal of epidermal stem cells, and ING5 knockdown causes differentiation of epidermal stem cells. Similar to other ING proteins, ING5 possesses a PHD (plant homeodomain) type zinc finger, an NCR (novel conserved region), a LZL (leucine zipper like) finger, and two NLS (nuclear localization sequence) motifs.
