Background InformationCREB-regulated transcription coactivator 1 (UniProt: Q6UUV9; also known as Mucoepidermoid carcinoma translocated protein 1;Transducer of regulated cAMP response element-binding protein 1;TORC-1;Transducer of CREB protein 1) is encoded by the CRTC1 (also known as MECT1; TORC1; WAMTP1; KIAA0616) gene (Gene ID 23373) in human. CRTC1 is a 67.3 kDa protein that activates transcription through binding cAMP response element (CRE) sites on DNA. It is highly expressed in adult and fetal brain and is localized to specific regions such as the prefrontal cortex and cerebellum. Very low expression is observed in heart, spleen, lung, skeletal muscle, salivary gland, ovary, and kidney. Phosphorylation/dephosphorylation states of Ser151 are required for regulating transduction of CREB activity. It is mainly localized in the cytoplasm when phosphorylated by salt-induced kinase (SIK) or AMPK and when sequestered by 14-3-3 proteins. It is translocated to the nucleus upon dephosphorylation on Ser151. CRTC1 is shown to mediate early beta-amyloid-induced transcriptional and memory deficits and overexpression of CRTC1 is shown to rescue amyloid-induced transcriptional and cognitive deficits. CRTC1 is reported to ameliorate hippocampal-dependent learning and memory deficits in APPSw,Ind mice by enhancing the expression of a specific subset of CCTC1 target genes. A chromosomal aberration involving CRTC1 has been reported in mucoepidermoid carcinomas, benign Warthin tumors and clear cell hidradenomas.
