      Background InformationOrigin recognition complex subunit 1 (UniProt: Q13415; also known as Replication control protein 1) is encoded by the ORC1 (also known as ORC1L, PARC1) gene (Gene ID: 4998) in human. Orc1 is a component of the ORC complex that binds origins of replication. It is the largest subunit of ORC and its expression is cell-cycle regulated. It begins to accumulate in mid-G1 phase and reaches a peak level at the G1/S boundary. Upon S phase entry, Orc1 is ubiquitinated and targeted for destruction and its levels decline significantly even when levels of other ORC subunits remain unchanged. Its levels increase when cells enter the mitotic phase. DNA-binding of Orc1 is shown to be ATP-dependent. The expression of Orc1 in human cells is reported to be regulated by E2F and the assembly of pre-replication complex at all origins is shown to depend on the E2F/Rb pathway. Orc1 contains a BAH domain (aa 45-171) that mediates its binding to demethylated histone H4 Lysine 20 (H4K20me2), which is enriched at replication origins. Mutations in ORC1 gene are known to cause Meier-Gorlin syndrome that is characterized by severe intrauterine and postnatal growth retardation and poor weight gain. (Ref.: Kara, N., et al. (2015). J.
