      BackgroundNFκB (Nuclear factor κB) is a transcription factor that plays crucial roles in gene regulation including those involved in immune and inflammatory responses. Active NFκB is most commonly composed of the heterodimer DNA binding subunits p50 and p65 (RelA). These subunits are members of a multigene family called the rel family. In addition to p50 and RelA, three other proteins are implicated in dimer formation leading to transcriptional activation, p52, c-rel and rel B. Inhibitors to this process exist including another member of the rel family IκBα which binds to c-rel, p65 and relB preventing DNA binding by their homo- or heterodimers. It has recently been shown that NFκB plays a role in the apoptotic process - when RelA, the gene that codes for an NFκB subunit, was knocked out, this lead to the death of mouse embryos through apoptosis of the liver cells. Similarly, it has been shown in a wide range of cells that NFκB which has been inactivated by IκBα were more sensitive to TNF-α-induced apoptosis than control cells. Evidence exists for NFκB playing both anti- and pro-apoptotic roles - it has been suggested that different combinations of subunits may result in different outcomes in terms of cell survival.
