Keratins are heteropolymeric structural proteins which form the intermediate filament. These filaments, along with actin microfilaments and microtubules, compose the cytoskeleton of epithelial cells. Mutations in this gene are associated with epidermolytic hyperkeratosis. This gene is located within a cluster of keratin family members on chromosome 17q21.

      DISEASE: SwissProt: P04264 # Defects in KRT1 are a cause of epidermolytic hyperkeratosis (EHK) [MIM:113800]; also known as bullous congenital ichthyosiform erythroderma (BIE). EHK is a hereditary skin disorder characterized by blistering and a marked thickening of the stratum corneum. & Defects in KRT1 are the cause of Curth-Macklin type ichthyosis hystrix (IHCM) [MIM:146590]. & Defects in KRT1 are a cause of nonepidermolytic palmoplantar keratoderma (NEPPK) [MIM:600962]; also known as Unna- Thost disease. & Defects in KRT1 are a cause of annular epidermolytic ichthyosis (AEI) [MIM:607602]; also known as cyclic ichthyosis with epidermolytic hyperkeratosis. AEI resembles clinical and histologic features of both epidermolytic hyperkeratosis and ichthyosis bullosa of Siemens. & Defects in KRT1 are the cause of keratosis palmoplantaris striata III (PPKS3) [MIM:607654]; also known as striate palmoplantar keratoderma III (SPPK3). PPKS3 is an autosomal dominant disorder affecting palm and sole skin where stratum corneum and epidermal layers are thickened. There is no involvement of non-palmoplantar skin, and both hair and nails are normal.
