      Entrez Gene Number
Entrez Gene SummaryThis gene was identified by its oncogenic transforming activity in cells. The encoded protein is a member of the serine/threonine protein kinase family. This kinase can activate both the MAP kinase and JNK kinase pathways. This kinase was shown to activate IkappaB kinases, and thus induce the nuclear production of NF-kappaB. This kinase was also found to promote the production of TNF-alpha and IL-2 during T lymphocyte activation. Studies of a similar gene in rat suggested the direct involvement of this kinase in the proteolysis of NF-kappaB1,p105 (NFKB1). This gene may also utilize a downstream in-frame translation start codon, and thus produce an isoform containing a shorter N-terminus. The shorter isoform has been shown to display weaker transforming activity.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P41279 # Able to activate NF-kappa-B 1 by stimulating proteasome- mediated proteolysis of NF-kappa-B 1/p105. Plays a role in the cell cycle. The longer form of cot has some transforming activity, although it is much weaker than the activated cot oncoprotein.
SIZE: 467 amino acids; 52925 Da
      SUBUNIT: Interacts with NFKB1/p105.
      SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Expressed in several normal tissues and human tumor-derived cell lines.
DEVELOPMENTAL STAGE: The 58 kDa form is activated specifically during the S and G2/M phases of the cell cycle.
PTM: Autophosphorylated. The longer form undergoes phosphorylation on Ser residues mainly, and the shorter form on both Ser and Thr residues.
SIMILARITY: SwissProt: P41279 ## Belongs to the protein kinase superfamily. STE Ser/Thr protein kinase family. MAP kinase kinase kinase subfamily. & Contains 1 protein kinase domain.
      Molecular Weight45.3kDa

Quality Assuranceroutinely evaluated by activation of MEK1 (14-420). The activated MEK1 was used to activate MAPKinase 2/Erk2, unactive. Subsequent phosphorylation of MBP (13-104) was performed.
