      Background InformationToll-like receptor 10 (UniProt: Q9BXR5; also known as CD290) is encoded by the TLR10 (also known as UNQ315/PRO358) gene (Gene ID: 81793) in human. Toll-like Receptors (TLRs) are transmembrane proteins that are expressed on various immune cells. The extracellular N-terminal region of TLRs recognizes specific pathogen components. At least 10 different members of TLR family have been identified in humans that detect different pathogen associated molecular patterns (PAMPs), including lipopolysaccharides, flagellin, bacterial CpG DNA, and viral RNA and DNA. Recognition of PAMPs by TLRs is considered as a key process for the induction of an inflammatory response. TLR10 is a single-pass type I membrane protein that is highly expressed in spleen, lymph node, thymus, and tonsil. Higher expressions have also been reported in promyelocytic HL-60 and in B-cell lines. It acts via MYD88 and TRAF6, leading to cytokine secretion and the inflammatory response. In TLR10-expressing cells, IkappaB alpha is shown to be consistently inhibited suggesting that TLR10 suppresses NF-kB signaling upstream of IkappaB alpha degradation. TLR10 is synthesized with a signal peptide (aa 1-19) that is cleaved off in the mature form, which contains an extracellular domain (aa 20-576), a transmembrane domain (aa 577-597), and a cytoplasmic domain (aa 598-811).
