      Background InformationTight junction protein ZO-2 (UniProt Q9Z0U1; also known as Tight junction protein 2, Zona occludens protein 2, Zonula occludens protein 2) is encoded by the Tjp2 (also known as Zo2) gene (Gene ID 21873) in murine species. The tight junction (TJ) constitutes the barrier between the apical and the basolateral domains of the plasma membrane. The assembly and permeability of this barrier are dependent on the zonula occludens (ZO) membrane-associated guanylate kinase (MAGUK) proteins ZO-1, -2, and -3 found at the submembranous domain of TJs in epithelia and endothelia. The 160 kDa ZO-2 contains three PDZ domains (PDZ1/a.a. 10-97, PDZ2/a.a. 287-365, PDZ3/a.a. 489-570) at its N-terminal end, followed by an SH3 domain (a.a. 584-649), a GUK/GK homology domain (a.a. 660-858), and a short PDZ-binding motif (TEL; a.a. 1165-1167) at the C-terminus. In addition to associating with various transmembrane junction proteins via its C-terminal PDZ-binding motif, ZO-2 is also involved in modulating gene expression and cell proliferation by interacting with a number of proteins in the nucleus. Evidence suggests that ZO-2 is also involved in stress response and cytoprotective mechanisms.
