      Background InformationMerkel cells are neuroendocrine cells found in skin that have synaptic contacts with somatosensory afferent terminals. Merkel cell carcinoma (MCC), a rare, but deadly neuroendocrine skin tumor that is caused by clonal integration of a Merkel cell polyomavirus (MCPyV) into these cells. Human polyomavirus is small nonenveloped double-stranded DNA viruses with their genome divided into early, late, and noncoding control regions. The early region encodes for large T (LT) and small T (sT) regulatory proteins and the late region comprises structural genes that produce viral capsid proteins VP1, VP2, and VP3. MCPyV-associated tumorigenesis is characterized and mediated by the sole expression of LT and sT antigens. The LT antigens contain several common motifs and domains that are shown to be important for facilitating the viral life cycle. The N-terminal end of LT (aa 1 70 aa) contains the DnaJ domain that consists of the CR1 (aa 13-17) motif followed by the HPDKGG hexapeptide sequence responsible for Hsc70 binding. The C-terminal portion of LT contains several critical elements that are essential for viral replication. It is reported that the T-antigen origin binding domain (OBD; aa 308-433) of LT is responsible for recognition and binding of a minimum 71-bp origin of replication in the noncoding regulatory region. It also contains a zinc-finger motif (aa 437-528) with in the core polyomavirus helicase/ ATPase domain that is shown to be essential for LT oligomerization and initiation of replication.
