      Applications
ApplicationAnti-Keratin Epithelial Antibody, clone AE3 is an antibody against Keratin Epithelial for use in WB, IH(P).
      Key Applications
      • Western Blotting
      • Immunohistochemistry (Paraffin)
Application NotesImmunohistochemistry on unfixed frozen sections or formalin-fixed paraffin-embedded tissue sections. For paraffin-embedded sections, antigen retrieval (HEIR or trypsin treatment) is strongly recommended.

Western blotting: Clone AE3 recognizes the 65 to 67, 64, 59, 58, 56 and 52 kDa human cytokeratins of the basic subfamily.

Optimal working dilutions must be determined by end user.

SpecificityRecognizes most of the basic (Type II) keratins, HMW cytokeratins. It is broadly reactive and stains positively almost all epithelia and their neoplasms. AE3 reacts with all layers of epidermis (basal layer and above). The antibody is an excellent marker for distinguishing carcinomas from non-epithelial tumours. It has been used for studying the immunolocalisation of epidermal keratins, tissue distribution of keratins, various diseases of epidermis, oral mucosa and other epithelia and other epithelia development. AE3 and AE1 (Catalog Number MAB1612) have been used extensively for distinguishing carcinoma from non-carcinoma.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P05787 # Together with KRT19, helps to link the contractile apparatus to dystrophin at the costameres of striated muscle.
SIZE: 483 amino acids; 53704 Da
SUBUNIT: Heterotetramer of two type I and two type II keratins. keratin-8 associates with keratin-18. Associates with KRT20. Interacts with HCV core protein and PNN. When associated with KRT19, interacts with DMD. Interacts with TCHP.
      SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Observed in muscle fibers accumulating in the costameres of myoplasm at the sarcolemma membrane in structures that contain dystrophin and spectrin. Expressed in gingival mucosa and hard palate of the oral cavity.
PTM: Phosphorylation on serine residues is enhanced during EGF stimulation and mitosis. Ser-74 phosphorylation plays an important role in keratin filament reorganization.
      DISEASE:SwissProt: P05787 # Defects in KRT8 are a cause of cryptogenic cirrhosis [MIM:215600].
SIMILARITY: SwissProt: P05787 ## Belongs to the intermediate filament family.
MISCELLANEOUS: There are two types of cytoskeletal and microfibrillar keratin: I (acidic; 40-55 kDa) and II (neutral to basic; 56-70 kDa).
