      Background InformationHomer protein homolog 1 (UniProt: Q86YM7; also known as Homer-1) is encoded by the HOMER1 (also known as SYN47) gene (Gene ID: 9456) in human. Homer1, 2, and 3 constitute a family of postsynaptic scaffold proteins that play a central role in calcium signaling. Homer1 is a widely studied member of this family, comprising both synaptic structure and mediating postsynaptic signaling transduction. It is a tetrameric protein that binds and cross-links cytoplasmic regions of various proteins. It contains a WH1 domain (aa 1-110) that interacts with the PPXXF motif in GRM1, GRM5, ITPR1, DNM3, RYR1, RYR2, SHANK1, and SHANK3. It also contains a C-terminal coiled-coil region (aa 181-352) region that mediate self-multimerization. By physically linking GRM1 and GRM5 with ER-associated ITPR1 receptors, Homer1 helps coupling of surface receptors to intracellular calcium release. Three isoforms of Homer1 have been described that are produced by alternative splicing. Isoform 1 is reported to regulate the trafficking and surface expression of GRM5. Isoform 3 acts as a natural dominant negative, in dynamic competition with constitutively expressed isoform 1 to regulate synaptic metabotropic glutamate function. Isoform 3 may also be involved in the structural changes that occur at synapses during long-lasting neuronal plasticity and development. Homer1 can negatively regulate T cell activation by inhibiting the calcineurin-NFAT pathway. It acts by competing with calcineurin/PPP3CA for NFAT protein binding, hence preventing NFAT activation by protein phosphatase 3 catalytic subunit (PPP3CA). (Ref.: Luo, P., et al. (2012). Neurochem.
