      Background InformationTumor necrosis factor receptor superfamily member 21 (UniProt: Q9EPU5; also known as Death receptor 6, DR6, CD358) is encoded by the Tnfrsf21 (also known as Dr6) gene (Gene ID: 94185) in murine species. DR6 is a single-pass type I membrane glycoprotein that is an orphan member of the TNF receptor superfamily. It controls cell death and differentiation in a cell autonomous manner in different cell types. It is ubiquitous expressed and higher expression has been observed in adult spleen, thymus, testis, prostate, ovary, small intestine, colon, brain, lung and kidney tissue. It is also detected in fetal brain, liver and lung. DR6 is expressed at low levels in proliferating progenitors in the spinal cord, but it is highly expressed by differentiating neurons within the spinal cord and adjacent dorsal root ganglia. Expressed by developing neurons as they differentiate and enter a pro-apoptotic state. DR6 promotes apoptosis, possibly via a pathway that involves the activation of NF-kappa-B and also promote apoptosis mediated by BAX and by the release of cytochrome c from the mitochondria into the cytoplasm. It also participates in neuronal apoptosis in response amyloid peptides derived from amyloid precursor protein. DR6 is reported to be a substrate for ADAM10 and -secretase. DR6 is shown to be involved in peripheral nervous system (PNS) myelination and DR6 knockout mice display precocious myelination in the PNS. DR6 is synthesized with a signal peptide (aa 1-41), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 42-349), a transmembrane domain (aa 350-370), and a cytoplasmic domain (aa 371-655). It contains four TNFR-Cys repeats and its death domain is localized to amino acids 415-498. (Ref.: Columbo, A., et al. (2018). EMBO J. 37: e97390).
