      Background InformationPeptidyl-prolyl cis-trans isomerase A (UniProt: EC:5.2.1.8, PPIase A, Cyclophilin A,CyPA, Cyclosporin A-binding protein, Rotamase A) is encoded by the PPIA (also known as CYPA) gene (Gene ID: 5478) in human. CyPA is a ubiquitous enzyme with peptidyl-prolyl cis-trans isomerase (PPIase) activity that catalyzes the cis-trans-isomerization of X-Pro peptide bonds. It binds to proteins and acts as a chaperone in protein folding. CyPA also displays immunosuppressant activity via inhibition of nuclear factor of activated T cells (NFAT) pathway. It is secreted by several cell types, including platelets, macrophages, and vascular endothelial cells. Its secretion also occurs in response to oxidative stress in vascular smooth muscle cells. Once released from cells, it binds to the extracellular matrix metalloproteinase (MMP) inhibitor CD147 expressed on plasma membrane of various cell types. Binding of extracellular CyPA to CD147 induces monocyte adhesion, migration, and MMP activity that can lead to extracellular matrix degradation. Acetylation of lysine 125 on CyPA is reported to inhibit the catalysis of cis- to trans- isomerization. This acetylation antagonizes the immunosuppressive effects of cyclosporin by blocking the sequential steps of cyclosporin binding and calcineurin inhibition. Two isoforms of CyPA have been described that are produced by alternative splicing. Clone 8H7 is reported to reduce monocyte/macrophage migration, both in vitro and in vivo, and also displays antithrombotic effects by blocking CyPA-dependent activation of platelets and thrombus formation.
