      • G-protein coupled receptor DEZ
Background InformationChemokine-like receptor 1 (UniProt P97468; also known as G-protein coupled receptor 27, G-protein coupled receptor DEZ) is encoded by the Cmklr1 (also known as Dez, Gpcr27, mcmklr1) gene (Gene ID 14747) in murine species. Cmklr1 is a 7-transmembrane G-protein-coupled receptor (GPCR) that is activated by chemoattractant chemerin. Chemerin is shown to induce the migration of macrophages and dendritic cells (DCs) in vitro, while in vivo studies using Cmklr1-knockout mice indicate an anti-inflammatory role of this receptor, possibly due to the recruitment of tolerogenic plasmacytoid DCs. In addtion, chemerin/Cmklr1 interaction is also reported to promote adipogenesis and angiogenesis. The anti-inflammatory lipid mediator, resolving E1, is another known Cmklr1 ligand that functions to prevent leukocyte infiltration and proinflammatory gene expression.

      Applications
ApplicationAnti-CMKLR1 Antibody, clone BZ194 is an antibody against CMKLR1 for use in Flow Cytometry.
      Key Applications
      • Flow Cytometry
Application NotesFlow Cytometry Analysis: A representative lot detected Cmklr1 expression on CD11c+/CD11b+/B220- myeloid dendritic cells (mDC) from the spinal cords of mice with acute experimental autoimmune encephalomyelitis (EAE) (Graham, K.L., et al. (2009). J. Immunol.183(10):6717–6723).
Flow Cytometry Analysis: A representative lot detected exogenously expressed mouse Cmklr1, but not human Cmklr1, on the respective murine pre-B lymphoma L1.2 transfectants (Zabel, B.A., et al. (2006). Exp. Hematol. 34(8):1106-1114).
Flow Cytometry Analysis: A representative lot detected Cmklr1 immunoreactivity among a subpopulation of CD11c+ mouse bone marrow cells, while no Cmklr1 immunoreactivity was detected among TCR-β-/CD19-/CD11c+ murine dendrditic cells (DCs) from bone marrow, spleen, lymph nodes, and blood (Zabel, B.A., et al. (2006). Exp. Hematol. 34(8):1106-1114).
Flow Cytometry Analysis: A representative lot detected Cmklr1 immunoreactivity associated with murine F4/80+/CD11+ macrophages from peritoneal cavity, pleural cavity, and peripheral blood (Zabel, B.A., et al. (2006). Exp. Hematol. 34(8):1106-1114).
      Biological Information
ImmunogenKLH-conjugated linear peptide corresponding to the N-terminal extracellular region of mouse CMKLR1.

SpecificityClone BZ194 specifically stains transfected L1.2 murine pre-B lymphoma cells that express mouse, but not human Cmklr1. Clone BZ194 does not stain murine cells known to express CXCR1 through 6 or CCR1 through 10 (Zabel, B.A., et al. (2006). Exp. Hematol. 34(8):1106-1114).
