DescriptionAnti-PRMT5 Antibody
OverviewPRMT5 (aka JBP1 Janus Binding Protein1) is a member of a family of enzymes that deposit methyl groups on arginine residues of proteins, including both histone and non-histone substrates.
Alternate Names
  • 72 kDa ICln-binding protein
  • HMT1 hnRNP methyltransferase-like 5
  • Jak-binding protein 1
  • SKB1 homolog
  • SKB1 homolog (S. pombe)
  • Shk1 kinase-binding protein 1 homolog
  • protein arginine methyltransferase 5
  • skb1 (S. pombe) homolog
Background InformationArginine methylation of proteins is mediated by the Protein-Arginine Methyl Transferase (PRMT) family of enzymes, which are important in signal transduction, transcription, RNA transport, and splicing. PRMTs are divided into two types defined by their activity. Type I PRMTs (including PRMT 1, 3, 4, and 6) are characterized by the formation of asymmetric dimethylated arginine residues. Type II PRMTs, which include only PRMT5, are defined by the formation of symmetric dimethylated arginine residues. Substrates for PRMT5 include myelin basic protein (MBP) and the spliceosomal D1 and D3 proteins. PRMT5 (also known as Skb1Hs/JBP1) exists as homooligomeric complexes, which includes a dimer and tetramer. Homooligomerization of PRMT5 has an important role in its ability to methylate the MBP protein.

UniProt SummaryFUNCTION: SwissProt: O14744 # Methylates specific arginine residues in the small nuclear ribonucleoproteins Sm D1 and Sm D3 to monomethylarginine and to symmetrical dimethylarginines (sDMAs). Methylates SUPT5H. Mono- and dimethylates arginine residues of myelin basic protein (MBP) in vitro. Plays a role in the assembly of snRNP core particles. May play a role in cytokine-activated transduction pathways. Negatively regulates cyclin E1 promoter activity and cellular proliferation. May regulate the SUPT5H transcriptional elongation properties. May be part of a pathway that is connected to a chloride current, possibly through cytoskeletal rearrangement. Methylates histone H2A/H4 'Arg-3' during germ cell development. Methylates histone H3 'Arg-8', which may repress transcription.
SIZE: 637 amino acids; 72684 Da
SUBUNIT: Forms, at least, homodimers and homotetramers. Interacts with PRDM1 (By similarity). Component of the methylosome, a 20S complex containing at least pICLn, PRMT1/SKB1 and MEP50. Component of a high molecular weight E2F-pocket protein complex, CERC (cyclin E1 repressor complex). Also interacts with Sm proteins, JAK2, SSTR1 and SUPT5H. Associates with SWI/SNF remodeling complexes containing SMARCA2 and SMARCA4.
SUBCELLULAR LOCATION: Cytoplasm.
TISSUE SPECIFICITY: Ubiquitous.
PTM: Disulfide bonds and non-covalent association mediate homooligomers formation.
SIMILARITY:SwissProt: O14744 ## Belongs to the protein arginine N-methyltransferase family.
