SpecificityThe breast cancer susceptibility gene, BRCA1*, is located on chromosome 17 (at q21), and is mutated in the majority of heritable breast cancers. The mutations within the coding region occur throughout the gene with little apparent clustering. The majority of BRCA1 mutations result in shortened BRCA1 gene products, creating a need for an amino-terminal marker. Antibody MAB4132 recognizes both the full-length and mutated (shortened) forms of BRCA1 in breast cancer cell lines. It also recognizes over-expressed protein in transiently transfected Cos cells. A single cross-reacting protein of approximately 27 kDa is also seen upon immunoprecipitation. No other cross-reacting proteins are detected.

BRAC1 is 220kDa.

The encoded protein combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as BASC for BRCA1-associated genome surveillance complex. This gene product associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complex. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in this gene are responsible for approximately 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Alternative splicing plays a role in modulating the subcellular localization and physiological function of this gene. Many alternatively spliced transcript variants have been described for this gene but only some have had their full-length natures identified.
