Background InformationIRAK1, also known as Interleukin-1 receptor associated kinase 1, belongs to the tyrosine kinase related (TKL) family and is involved with mRNA stabilization and activation of NF-kappa B. IRAK1 is located in all tissue as 2 isoforms, with 1 binding quickly and then degrading while isoform 2 is activated by kinase, resulting in a slower more sustained response to cytokines. Phosphorylation of IRAK1 leads to recruitment of the adapter protein PELI1. IRAK1 utilizes 2 pathways, the first one allows for mediation of IL-1 inflammatory responses when TRAF6 is bound, which activates NF-kappa B, thereby inhibiting I kappa B alpha. A second pathway does not involve TRAF6 and results in the stabilization of mRNA.

UniProt SummaryFunction: Binds to the IL-1 type I receptor following IL-1 engagement, triggering intracellular signaling cascades leading to transcriptional up-regulation and mRNA stabilization. Isoform 1 binds rapidly but is then degraded allowing isoform 2 to mediate a slower, more sustained response to the cytokine. Isoform 2 is inactive suggesting that the kinase activity of this enzyme is not required for IL-1 signaling. Once phosphorylated, IRAK1 recruits the adapter protein PELI1.
