Application CommentsThis antibody will block aggregation of cell lines mediated by ICAM-1 and blocks homotypic binding of purified populations of activated T- and B-lymphocytes and aggregation of mixed T- and B-cell blasts. Will inhibit T-cell adhesion to normal human endothelial cells. Activation induced by cell-cell contact (mixed lymphocyte reaction, T cell mediated B-cell activation) is significantly inhibited. This antibody will also block elements of both effector arms of the immune system (cytotoxic cell function and Ig production). This antibody has been reported to work for immunofluorescence and flow cytometry studies. Antibody should be titrated for optimal results in individual systems.

DescriptionPurified mouse monoclonal antibody generated by immunizing mice with the specified immunogen and fusing Balb/c splenocytes with p3-NS-1/Ag4-1 (NS1) mouse myeloma cells (see application references). Recognizes the ~85-115 kDa ICAM-1 protein.
BackgroundCell adhesion molecules are membrane glycoproteins that play a major role in inflammation and neoplastic disease. Four primary groups of adhesion molecules have been described, which includes the integrin receptor family, the immunoglobulin superfamily, selectins, and cadherins. The intercellular adhesion molecules (ICAM-1, ICAM-2, and ICAM-3) belong to the immunoglobulin superfamily that are the counter receptors for the leukocyte integrin, lymphocyte function associated antigen 1 (LFA-1, CD11a/CD18). ICAM-1 (CD54) is a single chain glycoprotein with 7 potential N-linked glycosylation sites. It is expressed in an inducible dependent manner by endothelial and epithelial cells as well as fibroblasts, macrophages, lymphocytes, and leukocytes. ICAM-1 can mediate adhesion by binding to CD11b/CD18 (Mac-1) enhancing antigen specific T-cell activation. ICAM-1 also binds to CD43 and to Plasmodium falciparum infected erythrocytes distinct from the LFA-1 binding site. Studies indicate that ICAM-1 may also be involved in the development of atherosclerotic plaques; increased expression of ICAM-1 has been observed in all types of atherosclerotic lesions except fibrous placques. It is thought that endothelial cell expression of ICAM-1 mediates the recruitment of leukocytes to the lesion as it is suggested by its role in inflammation.
