DescriptionAnti-19S regulator non-ATPase subunit S5a/Rpn10 Antibody, clone S5a-18
OverviewRpn10 (S5a) is a non-ATPase subunit of the 19S regulatory complex of the 26s proteasome. Subunit Rpn10 (S5a) of the 26S proteasome was originally identified as a protein capable of binding poly-ubiquitin chains of four or more ubiquitins, in the absence of other 26S proteasome subunits.

Entrez Gene Number
Entrez Gene SummaryThe 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of 2 complexes, a 20S core and a 19S regulator. The 20S core is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. The 19S regulator is composed of a base, which contains 6 ATPase subunits and 2 non-ATPase subunits, and a lid, which contains up to 10 non-ATPase subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway. An essential function of a modified proteasome, the immunoproteasome, is the processing of class I MHC peptides. This gene encodes one of the non-ATPase subunits of the 19S regulator lid. Pseudogenes have been identified on chromosomes 10 and 21.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P55036 # Binds and presumably selects ubiquitin-conjugates for destruction. Displays selectivity for longer polyubiquitin chains. Modulates intestinal fluid secretion.
SIZE: 377 amino acids; 40737 Da
SUBUNIT: The 26S proteasome is composed of a core protease, known as the 20S proteasome, capped at one or both ends by the 19S regulatory complex (RC). The RC is composed of at least 18 different subunits in two subcomplexes, the base and the lid, which form the portions proximal and distal to the 20S proteolytic core, respectively. Directly interacts with NUB1. Interacts with SQSTM1.
DOMAIN: SwissProt: P55036 The 2 UIM motifs are involved in the binding to a multi- ubiquitin chain in a cooperative way.
PTM: Phosphorylated upon DNA damage, probably by ATM or ATR.
SIMILARITY: Belongs to the proteasome subunit S5A family. & Contains 2 UIM (ubiquitin-interacting motif) repeats. & Contains 1 VWFA domain.
Molecular Weight41 kDa
