      Background InformationVesicular acetylcholine transporter (UniProt: Q62666; also known as VAChT, rVAT, Solute carrier family 18 member 3) is encoded by the Slc18a3 (also known as Vacht, Vat) gene (Gene ID: 60422) in rat. Acetylcholine (ACh) is synthesized in nerve terminals from choline and acetyl coenzyme A by the cytoplasmic choline acetyltransferase. ACh is then transported into synaptic vesicles where it is stored until release. This transport from cytoplasm into synaptic vesicles is achieved via vesicular acetylcholine transporter (VAChT) that acts as an antiporter to couple ACh entry into synaptic vesicles with proton efflux. VAChT is a mutli-pass membrane protein with seven cytoplasmic domains, twelve transmembrane domains, and six lumenal domains. High expression of VAChT is observed in all major cholinergic neurons, including postganglionic parasympathetic cells, preganglionic sympathetic and parasympathetic cells, ventral spinal cord, and brainstem motoneurons. It is weakly expressed in the cortex and hippocampus. Elevated levels of VAChT are reported to affect its own intraneuronal trafficking and can remodel striatal cholinergic interneuron morphology and reduce the number and increase the size of cholinergic varicosities.
