      Background InformationDNA topoisomerase 2-binding protein 1 (UniProt Q92547), also known as DNA topoisomerase II-binding protein 1, DNA topoisomerase II-beta-binding protein 1, TopBP1) is encoded by the TOPBP1 gene in human. TopBP1 plays crucial roles in the maintenance of genomic integrity by participating in DNA damage repair and cell cycle checkpoints regulation. Specifically, TopBP1 participates in the loading of Cdc45 for the assembly of the preinitiation complex necessary for chromosomal DNA replication. TopBP1 is also essential for the activation of cyclin E/CDK2 and Cdc7/Dbf4 kinases, as well as DNA polymerase chromatin loading. In response to replication fork stalling or DNA damage, TopBP1 mediates the activation of ATR-ATRIP complex, leading to the phosphorylation of ATR substrates, such as Chk1, Nbs1, Smc1, and H2AX. In addition, association of TopBP1 with Nbs1 is involved in homologous recombination repair upon double-stranded DNA break. TopBP1 also binds E2F1 and p53 and suppresses the pro-apoptotic activity of p53 and E2F1 in cancer cells. TopBP1 knockdown in 3T3 and murine embryonic fibroblasts induces senescence as a result of cellular DNA breakage and cell cycle arrest.
