      Background InformationInterleukin-9 receptor (UniProt: Q01114; also known as IL-9 receptor, IL-9R, CD129) is encoded by the Il9r gene (Gene ID: 16199) in murine species. IL-9R is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-37), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 38-270), a transmembrane domain (aa 271-291), and a cytoplasmic domain (aa 291-468). It contains a WSXWS motif (aa 244-248) that is essential for proper protein folding and cell surface receptor binding. It also contains a Box-1 motif (aa 301-309) that is required for JAK interaction and/or activation. IL-9R is composed of the IL-9R a-chain and common g-chain. IL-9R is expressed on mast cells, activated T cells, ILC2 cells, and basophils. It is also selectively induced on B memory cells after primary immunization and this expression is induced by stimulation with CD40. IL-9R expression on B memory cells is shown to be essential for a normal recall antibody response in murine models. Signaling by IL-9R is shown to result in the activation of major signaling pathways involving STAT1, STAT-3, and STAT-5, MAPK, and PI3K. IL-9R signaling is shown to be pro-inflammatory and it promotes allergic inflammation and airway remodeling in lungs. It is also important in cell growth and differentiation of T cells, B cells, mast cells, hematopoietic progenitors, epithelial cells, and airway smooth muscle cells. (Ref.: Takatsuka, S., et al.
