      Background InformationInsulin-like growth factor 1 receptor (EC:2.7.10.1; UniProt P08069; also known as CD221, IGF-I receptor, Insulin-like growth factor I receptor) is encoded by the IGF1R (also known as CD221, IGFIR, IGFR, JTK13) gene (Gene ID 3480) in human. The insulin-like growth factor 1 (IGF-1) receptor is a receptor tyrosine kinase activated by IGF-1 and IGF-2. IGF-1 receptor is initially produced as a precursor protein with a signal peptide sequence (aa 1-30). Post-translational cleavages produce one extracellular alpha (aa 31-736) chain and one transmembrane beta chain (aa 741 – 1367) linked together by a disulfide bond. Two sets of alpha-beta dimers then form the functional tetrameric receptor complex via a disulfide linkage between their alpha chain. Upon ligand binding, the alpha chains induce the tyrosine autophosphorylation of the beta chains, which in turn trigger cell type-specific downstream signaling cascade of events.
