Background Information
Tie-2 is a member of the tyrosine protein kinase family, is a transmembrane receptor for angiopoietin 1, and may represent one of the earliest markers for cell lineage within mammalian endothelial cells. Its expression is almost only found in human, mouse and rat endothelial cells and angioblasts. Most likely, Tie-2 regulates the differentiation, proper dispersal, and proliferation of these cells during angiogenesis. Mutations in TEK are a cause of VMCM or dominantly inherited venous malformations which are characterized by serpiginious, dilated veins. Tie-2 can be found in placenta and lung tissue. Lower levels of the protein can also be found in kidney, umbilical vein endothelial cells, and brain.

Recognizes human Tunica internal endothelial cell kinase (TIE-2), also described as Angiopoietin-1 receptor, p140 TEK, or tyrosine protein kinase receptor. The peptide sequence used as immunogen is 100% conserved in human, rat, mouse, and 94% in bovine.
FUNCTION: This protein is a protein tyrosine-kinase transmembrane receptor for angiopoietin 1. It may constitute the earliest mammalian endothelial cell lineage marker. Probably regulates endothelial cell proliferation, differentiation and guides the proper patterning of endothelial cells during blood vessel formation. CATALYTIC ACTIVITY: ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate. SUBUNIT STRUCTURE: Interacts with PTPRB. SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein. TISSUE SPECIFICITY: Predominantly expressed in endothelial cells and their progenitors, the angioblasts. Has been directly found in placenta and lung, with a lower level in umbilical vein endothelial cells, brain and kidney. PTM: Phosphorylated. Phosphorylation is induced by ANGPT1 and ANGPT2. ANGPT1-induced phosphorylation is impaired during hypoxia. Dephosphorylated by PTPRB. INVOLVEMENT IN DISEASE: Defects in TEK are a cause of dominantly inherited venous malformations (VMCM) [MIM:600195]; an error of vascular morphogenesis characterized by dilated, serpiginous channels. The expression of cytoplasmic and surface proteins in cultured human skin fibroblasts, human umbilical cord cells obtained after normal delivery on gestation week 38-40, and mesenchymal bone marrow stem cells was compared by the methods of immunocytochemistry and flow cytofluorometry. Bone marrow mesenchymal stem cells expressed a great variety of marker proteins typical of stem and progenitor cells and did not express proteins typical of differentiated cells. Fibroblast-like umbilical cord cells expressed markers of both stem cells and differentiated cells. Fibroblasts of dermal origin were characterized by intensive expression of proteins typical of differentiated cells.
