Application Comments
Antibody specificity was examined in the rat cortex, hypothalamus, raphe nuclei, and spinal cord. Staining is completely eliminated by pretreatment of antibody with Serotonin (5-HT) Transporter Control Peptide (Cat. No. PP87) at a concentration of 5 µg/ml. Low levels of glutaraldehyde (0.1%-0.3%) may be used in conjunction with paraformaldehyde for tissue perfusion fixation. The specificity of this antibody was also determined by immunoblotting analysis using rat brain extracts of the cortex, hypothalamus, midbrain, and hindbrain, which specifically detected a single band. Antibody should be titrated for optimal results in individual systems.

Description
Rabbit polyclonal antibody supplied as lyophilized, undiluted serum. Recognizes the serotonin (5-HT) transporter protein.
Background
The serotonin (5-hydroxytryptamine; 5-HT) transporter (5-HTT) plays an important role in the re-uptake of serotonin from the nerve terminal to ultimately terminate the serotonin signal. The 5-HTT plays a critical role in the regulation of serotonin neurotransmission, which has been linked to mood and personality disorders, anxiety, eating disorders, and insomnia. The predicted 12 transmembrane serotonin transporter contains potential sites for phosphorylation and glycosylation. Because serotonin is involved in such a variety of biological processes, the 5-HTT has been a target for the development of antidepressants and psychostimulants. Blocking serotonin uptake at the synapse has been achieved using structurally related chemicals called Selective Serotonin Re-uptake Inhibitors (SSRI) which alleviate disorders such as depression and alcoholism. The 5-HTT is localized to the plasma membrane of neurons and glial cells, as well as blood platelets, placenta, and endothelial cells.
