Leucine-rich repeat kinase 2 (LRRK2) is a 2,527 amino acid protein encoded by the PARK8 gene and belonging to the ROCO family of proteins. This protein family is characterized as having a unique interaction between the Roc domain and the intrinsic GTPase activity along with a COR domain. LRKK2 consists of 5 different domains; a tyrosine kinase-like MAPKKK-related domain, WD40 propeller motif repeats, a Roc (Ras-like small GTPase family) domain, a COR (C-terminal of Roc) domain, and an LRRK. LRRK2 is mainly localized to the cytoplasm and expression within tissue is most highly observed within the brain. It is found in lower levels within the liver, heart, lung, skeletal muscles, placenta, pancreas, and kidney. The exact function of LRRK2 is unknown; however mutations to any of the five domains are the most commonly known causes of Parkinson’s disease, both familial and sporadic. Further study of this protein may lead to a better understanding of the disease’s pathogenesis, as well as potential therapeutic markers.

This gene is a member of the leucine-rich repeat kinase family and encodes a protein with an ankryin repeat region, a leucine-rich repeat (LRR) domain, a kinase domain, a DFG-like motif, a RAS domain, a GTPase domain, a MLK-like domain, and a WD40 domain. The protein is present largely in the cytoplasm but also associates with the mitochondrial outer membrane. Mutations in this gene have been associated with Parkinson disease-8. [provided by RefSeq].
FUNCTION: Probable protein kinase whose role is not yet known. May play a role in the phosphorylation of proteins central to Parkinson disease. May also have GTPase activity.
CATALYTIC ACTIVITY: ATP + a protein = ADP + a phosphoprotein.
SUBUNIT STRUCTURE: Interacts with PARK2.
SUBCELLULAR LOCATION: Cytoplasm. Membrane; Peripheral membrane protein. Note: Localized in the cytoplasm and associated with cellular membrane structures. Associates with the mitochondrial outer membrane.
TISSUE SPECIFICTY: Expressed throughout the adult brain, but at a lower level than in heart and liver. Also expressed in placenta, lung, skeletal muscle, kidney and pancreas. In the brain, expressed in the cerebellum, cerebral cortex, medulla, spinal cord occipital pole, frontal lobe, temporal lobe and putamen. Expression is particularly high in brain dopaminoceptive areas.
INVOLVEMENT IN DISEASE: Defects in LRRK2 are the cause of Parkinson disease type 8 (PARK8) [MIM:607060]. A slowly progressive neurodegenerative disorder characterized by bradykinesia, rigidity, resting tremor, postural instability, neuronal loss in the substantia nigra, and the presence of neurofibrillary MAPT (tau)-positive and Lewy bodies in some patients.
