Anti-IKKβ Antibody: SDB (Sicherheitsdatenblätter), Analysenzertifikate und Qualitätszertifikate, Dossiers, Broschüren und andere verfügbare Dokumente.
IKKbeta (IkB Kinase, IKKbeta, I-Kappa-B kinase-beta) is a member of the IKK complex which is composed of IKKalpha, IKKbeta, IKKgamma and IKAP. IkB Kinase (IKK) complex is a critical component in NFkB regulation. In a majority of unstimulated cells, the NFkB transcription factors exist in their inactive form and are retained in the cytoplasm by the bound inhibitory IkB proteins. For NFkB to become activated, it must first disassociate from the inhibitor IkB. This occurs via the serine phosphorylation of IkB on Ser32 and Ser36. Post receptor stimulation, most notably by TNFalpha, NIK, a serine/threonine kinase, becomes activated and phosphorylates IKKalpha and IKKbeta. Phosphorylation of 2 sites at the activation loop of IKK beta is essential for activation of IKK by TNF and IL1. Once activated, IKKbeta autophosphorylates, which in turn decreases IKK activity and prevents prolonged activation of the inflammatory response. Additionally, IKKbeta activity can also be regulated by MEKK-1. IKK then binds to and phosphorylates IkBalpha on the two serine residues. This phosphorylation marks IkB for ubiquination and destruction by the proteosome. This free the NFkB complex and allows it to translocates to the nucleus.

Biological Information
Immunogen
Rabbit anti-IKKβ polyclonal antibody was raised against a peptide corresponding to amino acids 662 to 680 of human IKKβ (3,4), which differs from the corresponding murine sequence by one amino acid.
Concentration
Please refer to the Certificate of Analysis for the lot-specific concentration.
Host
Rabbit
Specificity
This antibody recognizes IKKbeta, does not cross reacts with IKKα or IKKγ.
NFKB1 (MIM 164011) or NFKB2 (MIM 164012) is bound to REL (MIM 164910), RELA (MIM 164014), or RELB (MIM 604758) to form the NFKB complex. The NFKB complex is inhibited by I-kappa-B proteins (NFKBIA, MIM 164008, or NFKBIB, MIM 604495), which inactivate NF-kappa-B by trapping it in the cytoplasm. Phosphorylation of serine residues on the I-kappa-B proteins by kinases (IKBKA, MIM 600664, or IKBKB) marks them for destruction via the ubiquitination pathway, thereby allowing activation of the NF-kappa-B complex. Activated NFKB complex translocates into the nucleus and binds DNA at kappa-B-binding motifs such as 5-prime GGGRNNYYCC 3-prime or 5-prime HGGARNYYCC 3-prime (where H is A, C, or T; R is an A or G purine; and Y is a C or T pyrimidine).[supplied by OMIM]
FUNCTION: Acts as part of the IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. Also phosphorylates NCOA3. CATALYTIC ACTIVITY: ATP + [I-kappa-B protein] = ADP + [I-kappa-B phosphoprotein]. SUBUNIT: Component of the I-kappa-B-kinase (IKK) core complex consisting of CHUK, IKBKB and IKBKG; probably four alpha/CHUK-beta/IKBKB dimers are associated with four gamma/IKBKG subunits. The IKK core complex seems to associate with regulatory or adapter proteins to form a IKK-signalosome holo-complex. Part of a complex composed of NCOA2, NCOA3, CHUK/IKKA, IKBKB, IKBKG and CREBBP. Part of a 70-90 kDa complex at least consisting of CHUK/IKKA, IKBKB, NFKBIA, RELA, IKBKAP and MAP3K14. Interacts with SQSTM1 through PRKCZ or PRKCI. Forms an NGF-induced complex with IKBKB, PRKCI and TRAF6. May interact with MAVS/IPS1. Interacts with NALP2. Interacts with TICAM1. Interacts with Yersinia yopJ. Interacts with FAF1; the interaction disrupts the IKK complex formation. Interacts with ATM. Part of a ternary complex consisting of TANK, IKBKB and IKBKG. Interacts with NIBP; the interaction is direct. SUBCELLULAR LOCATION: Cytoplasm. TISSUE SPECIFICITY: Highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis and peripheral blood. PTM: Upon cytokine stimulation, phosphorylated on Ser-177 and Ser-181 by MEKK1 and/or MAP3K14/NIK; which enhances activity. Once activated, autophosphorylates on the C-terminal serine cluster; which decreases activity and prevents prolonged activation of the inflammatory response. PTM: Yersinia yopJ may acetylate Ser/Thr residues, preventing phosphorylation and activation, which blocks the I-kappa-B signaling pathway. PTM: Ubiquitination on 'Ser-163' modulates phosphorylation on C-terminal serine residues. SIMILARITY: Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. I-kappa-B kinase subfamily. SIMILARITY: Contains 1 protein kinase domain.

Storage and Shipping Information
Storage Conditions
Stable at 2-8°C in undiluted aliquots for up to 12 months from date of receipt. Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C.
