Alternate Names
RB1-inducible coiled-coil protein 1
FAK family kinase-interacting protein of 200 kDa
Background Information
RB1-inducible coiled-coil protein 1 (UniProt: Q8TDY2; also known as FAK family kinase-interacting protein of 200 kDa, FIP200) is encoded by the RB1CC1 (also known as KIAA0203; RBICC) gene (Gene ID: 9821) in human. FIP200 plays an important role in several biological pathways including cell proliferation and autophagy. It regulates early and late events of autophagosome formation through direct interaction with Atg16L1. FIP200 expression has been shown in cytoplasm and in the nucleus. It serves as a positive regulator of the NF-kB and mTOR signaling. It also functions as a negative factor in FAK-Erk1/2 signaling pathway. It is reported to form a complex with p53 and/or hSNF5 and the large protein complex in mammalian cells and serves as a strong activator of retinoblastoma 1 (RB1), p16, and p21 promoters. The nuclear FIP200 is shown to enhance the RB1 pathway globally and behave as a tumor suppressor in some human malignancies. Its expression levels correlate well with those of RB1 in cancer cell lines and higher expression has been shown in human musculoskeletal and cultured osteosarcoma cells. Clone N1-8 is highly suited for immunohistochemistry analysis and it recognizes an epitope within amino acids 255-271. (Ref.: Hama, Y., et al. (2012). PLoS ONE 7(3); e32052).

Specificity
Clone N1-8 is a mouse monoclonal antibody that detects Focal adhesion kinase (FAK)-interacting protein of 200 kDa (FIP200). It targets an epitope within 17 amino acids from the N-terminal half.
Evaluated by Immunohistochemistry (Paraffin) in human breast cancer tissue sections.
Immunohistochemistry (Paraffin) Analysis: A 1:250 dilution of this antibody detected FIP200 in human breast cancer tissue sections.

RB1-inducible coiled-coil 1 (RB1CC1; also known as FIP200) plays important roles in several biological pathways such as cell proliferation and autophagy. Evaluation of RB1CC1 expression can provide useful clinical information on various cancers and neurodegenerative diseases. In order to realize the clinical applications, it is necessary to establish a stable supply of antibody and reproducible procedures for the laboratory examinations. In the present study, we have generated mouse monoclonal antibodies for RB1CC1, and four kinds of antibodies (N1-8, N1-216, N3-2, and N3-42) were found to be optimal for clinical applications such as ELISA and immunoblots and work as well as the pre-existing polyclonal antibodies. N1-8 monoclonal antibody provided the best recognition of RB1CC1 in the clinico-pathological examination of formalin-fixed paraffin-embedded tissues. These monoclonal antibodies will help to generate new opportunities in scientific examinations in biology and clinical medicine.
