DescriptionPurified mouse monoclonal antibody. Recognizes the ~60 kDa Src protein phosphorylated at Tyr418 (human, rat), Tyr423 (mouse), and Tyr416 (chicken, v-Src).
BackgroundSrc belongs to a family of 9 tyrosine kinases which regulate cellular responses to extracellular stimuli. Src is a non-receptor protein tyrosine kinase that transduces signals that are involved in the control of a variety of cellular processes such as proliferation, differentiation, motility, and adhesion. Src is normally maintained in an inactive state, but can be activated transiently during cellular events such as mitosis, or constitutively by abnormal events such as mutation (i.e. v-Src and some human cancers). Src becomes activated upon dephosphorylation of Tyr527 which leads to a conformational change of the Src molecule. This allows binding to other proteins via the SH2 and SH3 domain. Activated Src phosphorylates a variety of substrates involved in the mitogenic signaling pathway, cytoskeletal organization, cell-substrate adhesion, cell-cell adhesion, cell-cell communication and RNA processing.
