The subunit responsible for activation of adenylate cyclase. Binds to the ADP-ribosylation factor and modulates its activity. Catalyzes the ADP-ribosylation of the α-subunit of G-proteins, reducing intrinsic GTPase activity and activating the α-subunit.

Each lot of purified A subunit is 2-3 times more active than the intact toxin as an activator of adenylate cyclase (weight basis). In an ADP-ribosylation assay, the specific enzymatic activity of a typical lot of A subunit is approximately 2.0 times that of cholera toxin on a weight basis.

DescriptionCholera toxin is an oligomeric protein consisting of a single A subunit (M.W. 28,000) containing two polypeptide chains, A1 and A2, and a B subunit (M.W. 55,000) containing five polypeptide chains. Cholera toxin is the pathogenic agent responsible for the symptoms of cholera. Cholera toxin is also a potent activator of adenylate cyclase that binds the ADPribosylation factor and modulates its activity. The B subunit binds specifically to GM1 ganglioside receptors, which are expressed ubiquitously in mammalian cells, and assists in the transport of the A subunit through the membrane. The A subunit catalyzes the ADP-ribosylation of Gs which leads to the persistent activation of adenylate cyclase. The consequent increase in intracellular cAMP levels stimulates Cl- secretion and leads to severe dehydration. Cholera toxin is isolated from Vibrio cholerae type Inaba 569B by a modification of the methods of Rappaport, et al. and Mekalanos, et al. The purified A and B subunits are produced by a modification of the methods of Lai, et al.

SolubilityReconstitute with sterile distilled water. Please see vial label for reconstitution volume. Swirl gently during reconstitution as vigorous shaking or pipetting will result in damage to the toxin and a concomitant loss of biological activity. The purified A subunit is hydrophobic and vortexing may be required to get it into solution. DO NOT FREEZE solutions of cholera toxin subunits.
