A ubiquitous Ca2+-binding protein that serves as a physiological effector of a wide range of biological processes. Useful for the study of calmodulin-binding proteins immobilized on nitrocellulose, by Western blot or dot blot. Calmodulin purified from bovine brain and biotinylated.

Note: this product cannot be used for animal treatment, in vivo research, or any contact procedure with livestock.

Recommended reaction conditions
Protocol: Calmodulin, Biotinylated, Bovine Brain 10 ng of calcineurin were detected using 100 ng/ml of biotinylated calmodulin with the following method: 1. Immobilize the CaM-binding protein on nitrocellulose. 2. Prepare the reaction buffer: 50 mM Tris, pH 7.5, 200 mM NaCl, 500 µM CaCl2, 50 mM MgCl2, and 1% BSA. Block the nitrocellulose membrane by incubating at 20°C for 1 h with the reaction buffer. 3. Remove the blocking solution. Add the biotinylated calmodulin at 100 ng/ml in reaction buffer. Incubate at 20°C for 2 h. 4. Remove the biotinylated calmodulin solution. Wash 2 X 10 min in TBS/CaMg containing 0.05% TWEEN® 20, PROTEIN GRADE® Detergent, 10% Solution (Cat. No. 655206). 5. Incubate at 20°C for 30 min with Streptavidin, Alkaline Phosphatase Conjugated (Cat. No. 189732) diluted in the reaction buffer. 6. Wash 3 x 15 min with TBS/CaMg. Prepared substrate buffer: 100 mM Tris, 100 mM NaCl, pH 9.5, 50 mM MgCl2, and 0.5 mM CaCl2. Wash 1 x 15 min with substrate buffer. 7. Incubate for 20-30 min, protected from light, with NBT (330 mg/ml) and BCIP (0.165 mg/ml) in substrate buffer or use BCIP/NBT (Cat. No. 203790). 8. Wash extensively with H2O and air dry, protected from light.
