Background InformationMammalian reoviruses are nonenveloped viruses with a segmented, double-stranded RNA genome. Although these viruses can infect most mammalian species, they are usually asymptomatic in human. In neonatal SCID NOD mice they can cause lethal infection. Three major reovirus serotypes have been described that are differentiated by the capacity of antisera to neutralize viral infectivity and block hemagglutination. Type 1 and 3 sigma 1 contain receptor-binding domains in both the tail and head regions and in type 3 sigma 1 tail is shown to bind alpha-linked sialic acid. Sigma 1 is reported to engage glycan receptors and junctional adhesion molecule-A (JAM-A) and undergoes a conformational change during the proteolytic disassembly of virions to infectious subvirion particles (ISVPs) that accompany cell entry. Type 3 reovirus is shown to spread to the central nervous system and causes lethal encephalitis in murine models. Reovirus can also serve an oncolytic function and can hijack the activated Ras signaling pathway of host tumor cells. It can induce apoptosis in tumor cells through both extrinsic and intrinsic pathways. In this process viral uncoating is reported to be an essential factor.
