Background InformationMembrane-associated progesterone receptor component 1 (UniProt: O00264; also known as mPR, PGRMC1) is encoded by the PGRMC1 (also known as HPR6.6, PGRMC) gene (Gene ID: 10857) in human. PGRMC1 is a single pass homodimeric membrane protein that is widely expressed with highest expression observed in liver and kidney. It is also shown to be overexpressed in multiple types of cancers and can serve as an important biomarker of their proliferative state. It has short N-terminal extracellular domain (aa 1-24), a transmembrane domain (aa 25-43), and a cytoplasmic domain (aa 44-195). PGRMC1 is a component of a progesterone-binding protein complex with many cellular functions. It is required for the maintenance of uterine histoarchitecture and normal female reproductive lifespan. It also serves as an intracellular heme chaperone that regulates heme synthesis and acts as a heme donor for some hemoproteins. PGRMC1 contains a cytochrome b5 heme-binding domain (aa 72-171) that lacks the conserved iron-binding histidine residue at positions 107 and 131. PGRMC1 is reported to suppress p53 and Wnt/-catenin pathway to promote self-renewal of pluripotent stem cells (PSCs) and inhibit their differentiation.
