DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with mouse myeloma cells. Recognizes the ~76 kDa NSF-1 protein.
BackgroundSynaptic vesicle proteins can be divided into one of two classes; transport proteins involved in neurotransmitter uptake, and trafficking proteins which perform functions in vesicle docking, fusion, and budding. For example, synaptophysin may function in the process of neurotransmitter release, while synaptobrevin (VAMP) is believed to play a role in synaptic vesicle docking and exocytotic membrane fusion. Synaptobrevin has also been shown to bind synaptophysin with high specificity. Synapsins are highly abundant vesicle membrane proteins that are believed to regulate neurotransmitter release through phosphorylation-dependent interactions with cytoskeletal elements. Other proteins involved in vesicle exocytosis include syntaxin 1 and SNAP-25. In conjunction with synaptobrevin, these proteins form the SNARE (SNAP receptor) complex, which recruits other cytosolic factors including SNAP proteins (soluble NSF attachment proteins) and NSF (N-ethylmaleimide-sensitive factor). NSF acts in concert with SNAPs and SNAREs for the fusion of transport vesicles to their target membranes in synaptic transmission, intra-Golgi transport, endosome-to-endosome fusion and transcytotic vesicle-to-plasma membrane transport. In its functional form, NSF is a homotrimer of 76 kDa subunits. Each subunit contains an N-terminal domain and two homologous ATP binding domains (D1 and D2). Vesicle-to-target membrane docking occurs when the vesicle SNARE binds to its cognate target membrane SNARE. SNAPs α and γ then bind to this docking complex and mediate the binding of NSF and thus the formation of a 20 S fusion particle. It is thought that once NSF is bound, ATP hydrolysis by NSF initiates the fusion process.
