Background InformationMucin-13 (UniProt: Q9H3R2; also known as MUC-13, Down-regulated in colon cancer 1) is encoded by the MUC13 (also known as DRCC1, RECC, UNQ6194/PRO20221) gene (Gene ID: 56667) in human. Transmembrane mucins are glycoproteins involved in barrier function in epithelial tissues. Mucin-13 is a single-pass type I membrane protein but may also exist in a soluble form. It is highly expressed in epithelial tissues, particularly those of the gastrointestinal and respiratory tracts. Mucin-13 is characterized by a tandem repeat (TR) domain that is rich in serine and threonine residues that act as glycosylation sites. It is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 19-421), a transmembrane domain (aa 422-442), and a cytoplasmic domain (aa 443-512). The signal peptide targets Mucin-13 for translation into the endoplasmic reticulum where initial oligosaccharides are added, and the protein is transported to the Golgi apparatus for further glycosylation and posttranslational modifications. Mucin-13 contains three EGF-like domains (aa 173-211; 322-361; and 363-404) and one highly conserved SEA domain (aa 212-336) that contains a cleavage site. Through its EGF-like domain, it can enhance ErbB2/HER2 and ErbB3 signaling by increasing the localization of ErbB2 and ErbB3 to the cell membrane and by suppressing ligand-induced receptor internalization. Aberrant expression of Mucin-13 has been reported in a variety of epithelial carcinomas, including gastric, colorectal, and ovarian cancers. (Ref.: Maher, DM., et al. (2011). Mol.
