Background InformationInterleukin-23 subunit alpha (IL-23A or IL-23, p19 subunit) is a subunit of heterodimeric cytokine IL-23, which also contains the p40 subunit of interleukin 12 (IL-12B). The receptor of IL-23 is formed by the beta 1 subunit of IL-12 (IL-12RB1) and an IL-23 specific subunit, IL-23R. IL-23 activates the transcription activator STAT4, and stimulates the production of interferon-gamma. IL-23 is an important part of the inflammatory response against infection. It promotes upregulation of the matrix metalloprotease MMP9, increases angiogenesis and reduces CD8+ T-cell infiltration. Recently, IL-23 has been implicated in the development of cancerous tumors. In conjunction with IL-6 and TGF-β1, IL-23 stimulates naive CD4+ T cells to differentiate into a novel subset of cells called Th17 cells, which are distinct from the classical Th1 and Th2 cells. Th17 cells produce IL-17, a proinflammatory cytokine that enhances T cell priming and stimulates the production of proinflammatory molecules such as IL-1, IL-6, TNF-alpha, NOS-2, and chemokines resulting in inflammation.

SUBUNIT STRUCTURE: Heterodimer with IL12B; disulfide-linked. The heterodimer is known as interleukin IL-23.

SUBCELLULAR LOCATION: Secreted. Note: Secreted upon association with IL12B.

TISSUE SPECIFICTY: Secreted by activated dendritic and phagocytic cells and keratinocytes. Also expressed by dermal Langerhans cells (at protein level).

DEVELOPMENTAL STAGE: Expressed by newborns dendritic cells.

INDUCTION: Up-regulated by a wide array of pathogens and pathogen-products together with self-signals for danger or injury. Up-regulated in psoriatic dermal tissues, in dendritic cells of multiple sclerosis patients and in tumors.
