Brand Family Calbiochem®
Application Data
Detection of human acetylated histone H4 by immunoblotting. Samples: Acid-extracted proteins from HeLa cells treated with sodium butyrate at antibody concentrations of 0.5 µg/ml (lane 1) and 2 µg/ml (lane 2). Primary antibody: Anti-Histone H4, Acetylated (2-19) Rabbit pAb (Cat. No. 382160) (2 µg/ml). Detection: chemiluminescence.

Applications
Key Applications Immunoblotting (Western Blotting)
Immunocytochemistry
Immunoprecipitation
Application NotesImmunoblotting (0.5-2 µg/ml)
Immunocytochemistry (5-10 µg/ml)
Immunoprecipitation (5µg/500 µg lysate)
Application CommentsThe antibody detected acetylated histone H4 in acid-extracted proteins from HeLa cells. Cross-reacts with acetylated histone H2B from Tetrahymena and weakly cross-reacts with histone H2B from HeLa cells. May cross-react with other acetylated proteins. Using sodium butyrate, an inhibitor of histone deacetylases, enhances detection of acetylated histone H4. For immunocytochemistry, human A431 cells were treated with 5 mM sodium butyrate, an inhibitor of histone deacetylases, for 24 h and fixed with ice-cold 1% paraformaldehyde in PBS. Variables associated with assay conditions will dictate the optimal dilution.

DescriptionProtein A purified rabbit polyclonal antibody. Recognizes the ~10 kDa acetylated histone H4 protein.
BackgroundThe acetylation of core histones regulates gene expression through alterations in the chromatin structure. Lysine acetylation in the amino-terminal domains of histones H3 and H4 can alter the topology of the DNA in chromatin and probably influence its higher-order folding.
