BackgroundThe regulation of eukaryotic gene expression is marked by post-translational modification of nucleosomal histones, a key event that converts regions of chromosomes into transcriptionally active or inactive chromatin. A single nucleosomal core particle contains an octamer of histone proteins H2A, H2B, H3 and H4. Acetylated histone proteins confer accessibility of the DNA template to the transcriptional machinery. There are at least eight cloned human histone deacetylases. Studies have pointed to the possible involvement of histone deacetylases in human cancer. Histone Deacetylase 1 has been shown to interact directly with transcription factors and to deacetylate histone proteins H3 and H4. Studies have also shown that mammalian histone deacetylase (HDAC)-1, -2, and -3 are all capable of down-regulating p53 function. p53, the most commonly mutated gene in cancer cells, directs cell cycle arrest or induces programmed cell death (apoptosis) in response to stress. HDACs are classified by their sequence homology to the yeast HDACs. Class I proteins are related to Rpd3 and members of class II resemble HdaIp.
