Background InformationEstradiol is a major female sex hormone that regulates estrous and menstrual cycle. It is also present in smaller quantities in males where it is an active metabolic product of testosterone. In females, it is mainly produced by the granulosa cells in the follicles of ovaries. Smaller amounts of estradiol are also produced by the adrenal glands. It acts as an agonist for the estrogen receptor (ER) and can bind to both ERa and ERb. It is synthesized from cholesterol where androstenedione is a key intermediate which is converted to estradiol by aromatase. It is metabolized via hydroxylation into catechol estrogens. In the liver it can undergo hydroxylation into 2-hydoxyestradiol by CYP1A2, CYP3A4, and CYP2C9. Estradiol is reported to directly act on the central nervous system to modulate energy balance and reduced levels of estradiol following ovarian insufficiency have been linked to menopause, hyperphagia, and weight gain.
