DescriptionRabbit polyclonal antibody supplied as undiluted serum. Recognizes the ~8.6 kDa unconjugated and various forms of conjugated ubiquitin.
BackgroundUbiquitin is a highly conserved protein that functions in intracellular regulation. The ubiquitin system is the major pathway for selective protein degradation and is responsible for the degradation of abnormal and short-lived proteins. In the first step, ubiquitin is activated in an ATP-dependent reaction by ubiquitin-activating enzyme (E1). Activated ubiquitin is transferred to a ubiquitin conjugating enzyme (E2), then transferred to a protein substrate with the aid of ubiquitin-protein ligase (E3). An isopeptide linkage is formed between the C-terminus of the ubiquitin and an internal lysine residue of the substrate protein. Degradation requires the presence of a multi-ubiquitin chain on a substrate, which targets the protein for degradation by the 26S proteasome. Ubiquitin molecules are then released and recycled.
