Background InformationInterleukin-10 (UniProt: P22301; also known as IL-10, Cytokine synthesis inhibitory factor, CSIF) is encoded by the IL10 gene (Gene ID: 3586) in human. IL-10 is a homodimeric secreted anti-inflammatory cytokine produced by a variety of cells, including T-cells, macrophages, and mast cells. It is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature form. IL-10 acts on many cells of the immune system to limit excessive tissue disruption caused by inflammation. During infection it inhibits the activity of Th1 cells, NK cells, and macrophages, all of which are required for optimal pathogen clearance. It ameliorates the excessive Th1 and CD8+ T cell responses that are responsible for much of the immunopathology associated with infections. It also inhibits the release of pro-inflammatory cytokines. It is shown to control the inflammatory response of macrophages by reprogramming essential metabolic pathways including mTOR signaling. IL-10 binds to its heterotetrameric receptor comprising IL10RA and IL10RB, which leads to JAK1 and STAT2-mediated phosphorylation of STAT3 that in turn, STAT3 translocates to the nucleus where it drives expression of anti-inflammatory mediators.
