Recommended reaction conditions
Assay Protocol for Protease Activity 1. Dissolve 50 mg AZOCOLL® in 5 ml of buffer solution (e.g., PBS, pH 7.0) containing the proteolytic enzyme. 2. Incubate in stoppered tubes for 15 min at 37°C, agitating gently at 5 min intervals. 3. Cool the tubes in ice water and filter the contents through Whatman No. 1 filter paper. 4. Measure the absorbance at 520 nm, subtracting the blank value. 5. Plot absorbance versus protease activity. A linear relationship exists between protease concentration and the rate of dye liberation. Note: The rate of color yield from AZOCOLL® will vary with the individual proteases tested. This protocol is meant to serve as a guideline and may need to be modified for specific applications.
