Background InformationNEDD8 ultimate buster 1 (UniProt Q9Y5A7; also known as Negative regulator of ubiquitin-like proteins 1, Renal carcinoma antigen NY-REN-18) is encoded by the NUB1 (also known as BS4, NUB1L, NYREN18) gene (Gene ID 51667) in human. The Long (NUB1L) and Short spliced isoforms of NUB1 are interferon-inducible proteins that mediate the degradation of the ubiquitin-like protein NEDD8 and its protein conjugates. In addition, NUB1L can also facilitate the proteasomal degradation of another Ub-like protein FAT10. Cyclin E, p27, and p53 are among the proteins known to be targeted by NUB1. The ternary complex composed of UFD1, NPL4, and the AAA-ATPase P97/VCP (valosin-containing protein) plays a key role in recruiting and delivering ubiquitinated substrates to the proteasome. NUB1L is reported to interact with the complex and mediate NEDD8 proteasomal delivery.
