Application NotesImmunoprecipitation: A representative lot of this antibody clone precipitated PAGE bands of 66, 76, 120, 135 and 200 kDa (reduced) or 66, 110, 125, 130 and 190 kDa (non-reduced) from surface-biotinylated SCC9 cells.

Immunohistochemistry: A representative lot of this antibody clone was used in immunohistochemistry (acetone-fixed frozen human epithelial tissues including tongue, foreskin and ovary. Not for use on formalin-fixed tissue).

Flow Cytometry: A representative lot of this antibody clone stained human squamous cell carcinoma (SCC9), ovarian carcinoma (SKOV-3) and umbilical vein endothelium (HUVEC). Stains RKO or K562 cells transfected for CD104 expression. Does not stain monocytes, neutrophils, lymphocytes, T cells, B cells or platelets.

Functional Activity Assay: A representative lot of this antibody clone induced a sub-basal split at the basement membrane in non-wounded tissue. In addition, wound closure was significantly inhibited by ASC-8, as the epithelial tongue only covered 40 percent of the wound area at 120 hours post-wounding. These results demonstrate that the β4 adhesion-blocking antibody may have adverse effects on normal tissue (Egles, C., et al., 2010).

Entrez Gene SummaryIntegrins are heterodimers comprised of alpha and beta subunits, that are noncovalently associated transmembrane glycoprotein receptors. Different combinations of alpha and beta polypeptides form complexes that vary in their ligand-binding specificities. Integrins mediate cell-matrix or cell-cell adhesion, and transduced signals that regulate gene expression and cell growth. This gene encodes the integrin beta 4 subunit, a receptor for the laminins. This subunit tends to associate with alpha 6 subunit and is likely to play a pivotal role in the biology of invasive carcinoma. Mutations in this gene are associated with epidermolysis bullosa with pyloric atresia. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.

EB-PA is characterized by mucocutaneous fragility and gastrointestinal atresia, which most commonly affects the pylorus. & Defects in ITGB4 are a cause of generalized atrophic benign epidermolysis bullosa (GABEB) [MIM:226650]. This nonlethal form of junctional epidermolysis bullosa is characterized by life- long blistering of the skin, associated with hair and tooth abnormalities.
