DescriptionRabbit polyclonal antibody supplied as diluted serum. Recognizes the ~75 kDa and the ~50 kDa forms of GLT-1.
BackgroundPrecise regulation of excitatory amino acids (EAA) such as glutamate is essential, as glutamate is not only the major excitatory neurotransmitter, but also a potent excitotoxin. Defects in functional glutamate transport have been identified in the amyotrophic lateral sclerosis (ALS) brain and spinal cord, suggesting that defective glutamate transport may contribute to motor neuron degeneration. Three glutamate transporter subtypes have been identified in mammals, which include GLAST, GLT-1 and EAAC1. More recently, the homologous proteins EAAT1, EAAT2, and EAAT3 have been characterized in humans, respectively. EAAT4 and EAAT5 transporters have been localized to Purkinje cell synapses, and retina, respectively. The selective loss of glutamate transport in ALS appears to be specific for the GLT-1 (EAAT2) transporter subtyp. While GLT-1, GLAST and EAAC1 share approximately 50% sequence homology, the sequence used as the immunogen is unique to GLT-1.
