Application CommentsThe full-length sequence for BDNF can be found under accession number NP_001700. This antibody has been selected for its ability to neutralize the biological activity of human recombinant BDNF. The exact concentration of antibody required to neutralize human recombinant BDNF activity is dependent on the cytokine concentration, cell type, growth conditions, and the type of activity studied. Suggested neutralization concentration required to yield one-half maximal inhibition of BDNF activity is approximately 5-15 µg/ml in the presence of 2.5 ng/ml of human recombinant BDNF, in a neuron survival assay using embryonic chick dorsal root ganglia neurons. The detection limit for human recombinant BDNF is ~300 ng/lane under non-reducing and reducing conditions. The detection for human recombinant BDNF is ~12.5 ng/ml. Based on immunoblotting and ELISA, this antibody exhibits less than 2% cross-reactivity with human recombinant β-NGF, human recombinant NT-3, and human recombinant NT-4. Antibody should be titrated for optimal results in individual systems.

BackgroundNeurotrophic factors define a family of related proteins, which are important in the development, maintenance, and regulation of neurons in both the peripheral and central nervous system. In addition to the prototypical nerve growth factor (NGF), several other factors have been identified which interact with a family of tyrosine kinase receptors. TrkA is the kinase signaling receptor for NGF, while TrkB mediates signaling for both brain-derived neurotrophic factor (BDNF) and neurotrophin-4/5 (NT-4/5). TrkC is the primary receptor for neurotrophin-3 (NT-3), although this neurotrophin can also activate TrkA and TrkB receptors under certain conditions. Axotomy-induced cell death is one experimental paradigm that has been used to discriminate between the effects of various neurotrophic factors on the survival of different neuronal populations. BDNF, NT-3, and NT-4/5 act as motor neuron survival factors, whereas NGF has no effect on the survival of spinal cord motor neurons. NGF is neuroprotective for cholinergic neurons in the central nervous system. Retrogradely transported NGF-TrkA has also been shown to signal the CREB transcription factor in sympathetic neurons. A putative new member of this gene family, neurotrophin-6 (NT-6), has also been identified from the fish Xephophorus. Like NGF, NT-6 promotes the survival of sympathetic and sensory neurons, albeit with a lower potency.
