Background InformationApelin receptor (UniProt: P35414; also known as Angiotensin receptor-like 1, G-protein coupled receptor APJ, G-protein coupled receptor HG11) is encoded by the APLNR (also known as AGTRL1, APJ) gene (Gene ID: 187) in human. Apelin receptor(AR), a monomeric transmembrane protein, is a class A GPCR that is widely distributed in human. It is also expressed in embryonic and induced pluripotent stem cells. It is activated by its cognate peptide ligand, apelin, and induces a wide variety of effects, including vasoconstriction and dilation, strengthening cardiac muscle contractility, angiogenesis, fluid homeostasis, and energy metabolism. It also plays role in heart morphogenesis and promotes angioblast migration towards embryonic midline. Apelin-AR signaling has cardioprotective effects and administration of apelin-13 to glucose-deprived cultured cardiomyocytes has inhibitory effects on apoptosis. Glutamic acid 20 and aspartic acid 23 at the extracellular N-terminus of AR are reported to be the first physiological sites that bind apelin. After exposure to apelin, the receptor is internalized from the cell surface into an endosomal recycling compartment, from where it is recycled to the cell membrane. (Ref.: Hu, G., et al. (2021).
