What are the specifications of the SCX resin in the ZipTipSCX Pipette Tips?SCX resin is 12 mm beads with a 300 A pore size.
What is the binding capacity of ZipTipSCX?Binding capacity of ZipTipSCX is about 200-250 ng.
The mass spectra results indicate loss of peptides after purifying my sample with ZipTip-SCX. What do you suggest to improve my recoveries?We recommend pre-washing the tips with the elution solution (5% NH4OH, 30% MeOH) priop to equilibration with washing solution (0.1% TFA). This improves binding.

In addition, peptides should be bound under acidic conditions and eluted under basic conditions. It is recommended that you check the pH of your binding colution with pH paper to ensure it is acidic.

It may be that the eluted peptides were not allowed to dry completely before the matrix was added to the MALDI plate.
Can I use ZipTipSCX to purify samples from solutions containing salts?Yes, we have seen efficient binding from solutions containing up to 0.5M NaCl under acdic conditions. If the salt solution is basic binding will not occur.
When is it recommended to use ZipTipC18 versus ZipTip SCX?For all applications involving the desalting and concentration of peptides and proteins from aqueous solutions we recommend the ZipTipC18. If the peptide solutions caontains solvent or detergent we recommend the ZipTipSCX.
Can I use ZipTip SCX to remove glycerol from my sample?Gylcerol will interfere with mass spectroscopy. ZipTipSCX can remove up to 20-30% glycerol from a sample.
My spots on the MALDI plate are yellow after purifying my sample with ZipTip SCX. What could this be due to?This yellow color is due to the peptide sample was not fully dried on the MALDI plate before the matrix was applied. It is important for the sample to dry completely before the matrix is applied or you may not see any signal.
Can I use higher than 50% ACN to elute my sample from ZipTipC18 and ZipTipm-C18?Yes, however the tip should be prewet 5 times with 10 ml of 100% ACN followed by a wash of 10 ml 0.1% TFA. Then bind, wash and elute.
After elution from ZipTip my spots won't dry. Want can I do?For the initial prewet step wash the tip 5 times with 100% ACN followed by 0.1% TFA. Then bind, wash and elute as normal.
Are ZipTip pipette tips compatible with robotic / automated sample preparation systems?ZipTipm-C18 compatibility has been demonstarted with the following robotic / automated sample preparation systems:
